چکیده

Introduction Bluetongue virus (BTV) first emerged in the European Union (EU) in 2006, peaking at 45,000 cases in 2008. The EU spent million of euros in 2008 and 2009 on eradicating and monitoring programs, co-financed with member states. The number of cases in 2009 was 1118, with only 120 reported across the EU so far last year. Vaccination has proven itself as the most effective tool to control and prevent the disease and to facilitate the safe trade of live animals. Mammalian cells are commonly used as a substrate for production of most of the viral vaccines. BHK-21 cells are used for the production of bluetongue vaccines. Most companies use roller bottles or conventional bioreactors, but taking into account that the manufacturing cost is very important, the possibility of using SingleUse Bioreactor (SUB) technology as an alternative to roller bottles or conventional bioreactors was explored. Advantages like yields of production, time reduction (elimination of cleaning and sterilization steps needed for bioreactors, no validation process, etc) and quality of antigen production were studied.

Comparative study of bluetongue virus serotype 8 production on BHK-21 cells in a 50L Biostat® STR single-use bioreactors vs roller bottles

Background Bluetongue is a major disease of ruminant livestock that can have a substantial impact on income and animal welfare. Bluetongue virus serotype 8 (BTV-8) first emerged in the European Union in 2006, peaking at 45,000 cases in 2008. Zoetis (formerly Pfizer Animal Health) licensed bluetongue vaccines (Zulvac 4 Ovis, Zulvac 1 Ovis, Zulvac 1 Bovis, Zulvac 8 Ovis and Zulvac 8 Bovis and com...

Improved efficacy of a licensed acellular pertussis vaccine, reformulated in an adjuvant emulsion of liposomes in oil, in a murine model.

The immunogenicities and efficacies of a licensed diphtheria, tetanus, acellular pertussis, and inactivated poliovirus vaccine and the same vaccine formulated in a liposome/oil emulsion adjuvant were compared in a mouse model of pertussis respiratory infection. A single dose of the liposome/oil emulsion-adjuvanted vaccine produced significantly higher antibody levels than one dose of the licens...

Determination of Antibody Response against Inactivated Agalactia Vaccine in Small Ruminants

An inactivated agalactia vaccine was prepared with Mycoplasma agalactiae in fermenter. The culture was inactivated and subsequently adjuvanted with aluminum hydroxide gel. Four sheep and four goats were immunized with the experimental vaccine. Two doses of 1ml of vaccine were administered three weeks interval. Monthly, blood samples were taken from the vaccinated animals up to the fifth month...

Preparation of agalactia vaccine in fermentor

Mycoplasma agalactia was cultured in PPLO broth medium supplemented with sterilized normal horse serum in fermentor. Cultivation process continued at 37 ºC under aerobic conditions and growth was terminated after 26 hours cultured period with 2.2×109 Mycoplasma per ml. Samples were taken at different stages of growth. Culture was inactivated with 0.04 %v/v formaldehyde and the vaccine was adjuv...

Efficacy of inactivated velogenic Newcastle disease virus genotype VII ‎vaccine in broiler chickens

Newcastle disease (ND) causes severe economic losses in poultry production. Despite the intensive vaccination regimes of NDV in Egypt, many outbreaks are being reported. The present study focused on the preparation and evaluation of inactivated velogenic Newcastle disease virus vaccine (genotype VII) isolated from Egyptian broiler chicken during 2015-2016. Fifty-five tissue samples including tr...
